What Is Batch-Specific Testing?
Batch-specific testing means every production batch of a peptide is independently analyzed and receives its own Certificate of Analysis with unique data. This contrasts with generic or representative COAs, where a single set of test results is applied across multiple batches — sometimes for months or years.
The distinction matters because peptide synthesis is a chemical process with inherent variability. Raw material quality, equipment calibration, environmental conditions, and operator technique all introduce batch-to-batch variation that can only be detected through individual batch testing.
Why Batches Vary
Synthesis efficiency: SPPS coupling efficiency can vary between runs. A 99.5% coupling efficiency per step sounds high, but over 15 coupling cycles (for a 15-amino acid peptide), it means only 92.7% of chains are full-length. Small changes in coupling efficiency compound across the synthesis.
Purification outcomes: HPLC purification depends on column condition, solvent quality, gradient precision, and sample loading. The same crude peptide can yield different purities depending on purification parameters.
Lyophilization conditions: Freezing rate, shelf temperature during drying, and chamber vacuum all affect the final product's moisture content, cake appearance, and reconstitution behavior.
Generic vs. Batch-Specific COAs
A generic COA typically shows suspiciously round purity numbers (exactly 99.0%, for example), lacks a unique lot or batch number, and may use the same chromatogram image across products or time periods. It represents what the supplier believes their product should be, not what this specific batch actually is.
A batch-specific COA includes a unique lot number traceable to a specific production run, an actual HPLC chromatogram from that batch's analysis, a purity percentage calculated from that specific chromatogram, and ideally mass spectrometry data confirming molecular identity for that batch.
Impact on Research
Using peptides without batch-specific quality verification introduces an uncontrolled variable into your research. If your results change between experiments, is it due to a biological variable or a change in peptide quality? Without batch-specific data, you cannot answer this question.
For published research, reviewers and readers increasingly expect materials sections to include supplier name, catalog number, lot number, and stated purity. Batch-specific testing provides this essential provenance information.
What to Look For
When evaluating a supplier, ask these questions: Does every batch receive its own COA? Can you access the COA before purchasing? Does the COA include actual analytical data (chromatograms, spectra) or just numbers? Is the lot number on the COA traceable to the product you receive? A yes to all four indicates a supplier committed to batch-specific quality verification.
